6例健康志愿者,年龄23~33岁,体重46~71kg.单剂量口服氨茶碱控释片300mg,先于晨8h 服药(Ⅰ),取血时间为服药后2、4、5、6、9、12、16和24h.同时用荧光偏振免疫法(TDX)及紫外分光光度法(UV—3000)测定血中茶碱浓度.两周后改为晚8h 服药(Ⅱ),其它均相同。按单剂量口服一室开放模型计算,结果是:吸收速率常数和表现分布容积未见显著差异(P>0.05).荧光偏振免疫法和紫外分光光度法测血中茶碱浓度相关性较好(r=0.93,p<0.01).平均血浓度误差0.733±0.078μg/ml.
6 healthy volunteers, aged 23 to 33 years old, weighing 46 to 71 kg. A single dose of oral aminophylline controlled-release tablets 300mg, 8h before taking the drug (Ⅰ), taking blood time after taking 2,4,5,6 , 9, 12, 16 and 24h respectively.At the same time, the concentration of theophylline in serum was determined by fluorescence polarization immunoassay (TDX) and ultraviolet spectrophotometry (UV-3000) . The results of single-dose oral single-chamber open model calculation showed that there was no significant difference in absorption rate constants and volume of distribution (P> 0.05), fluorescence polarization immunoassay and UV spectrophotometry had a good correlation with theophylline concentration r = 0.93, p <0.01) .The mean blood concentration error was 0.733 ± 0.078μg / ml.
