目的 复制小鼠Lewis肺癌模型,检测威麦宁的抑瘤率。方法 采用流式细胞术分析、免疫组化法检测药物对模型鼠肿瘤细胞周期及对CyclinD1表达的影响。结果 威麦宁浓度在 1 0 0、1 5 0、2 0 0、2 5 0mg/(kg·d)时,对小鼠Lewis肺癌的抑制率分别为 1 9 1 4 %、33 5 9%、4 0 6 3%、5 1 5 6 %,药物对肿瘤的抑制作用具有剂量依赖性;药物可将肿瘤细胞阻滞在G0 ~G1期,并降低肿瘤细胞CyclinD1的表达(P <0 0 1)。结论 威麦宁可能是通过降低肿瘤细胞CyclinD1的表达,将肿瘤细胞阻止于G0 ~G1期,使细胞不能进入S期进行DNA复制,从而抑制肿瘤的生长。
Objective To replicate mouse Lewis lung cancer model and detect the inhibitory effect of Weimaining. Methods Flow cytometry and immunohistochemistry were used to detect the effects of drugs on the cell cycle and the expression of CyclinD1 in model mice. RESULTS: The inhibition rates of Lewis lung cancer in Wilmstine at doses of 100, 150, 200, and 250 mg/(kg·d) were 1914% and 33.59%, respectively. 4 0 6 3%, 5 1 5 6 %, the inhibitory effect of drugs on tumors was dose-dependent; drugs could arrest tumor cells in the G0 ~ G1 phase and reduce the expression of CyclinD1 in tumor cells (P <0 01). . Conclusion Weimaining may reduce the expression of CyclinD1 in tumor cells and block the tumor cells in G0-G1 phase, so that cells can not enter S phase for DNA replication, thus inhibiting tumor growth.
