目的探讨人与猪胆总管几何形态及顺应性的关系,为猪到人异种肝移植提供理论依据。方法取5例成年人尸体和50例3~12月龄湖北白猪的胆总管,在软组织生物力学实验机上测定胆总管的压力-直径关系,计算出顺应性。横断取材,冷冻切片,HE染色,用计算机图像分析系统测量其管径和管壁厚度。结果3~6月龄猪胆总管的管壁比成人的薄,其直径比成人的小(P<0.01),7~10月龄猪胆总管的壁厚和直径与成人的相近(P>0.05)。3~6月龄及11、12月龄猪胆总管的顺应性明显小于成人(P<0.01),7~10月龄猪胆总管的顺应性与成人的相近(P>0.05)。结论7~10月龄猪胆总管的几何形态和顺应性与成人的相近,从生物力学方面考虑,7~10月龄的猪肝作为人异种移植肝供源是可能的。
Objective To investigate the relationship between human and pig common bile duct geometry and compliance, and to provide a theoretical basis for xenotransplantation of pigs to humans. Methods The bile duct of 5 adult cadavers and 50 white pigs from 3 to 12 months of age were collected. The pressure - diameter relationship of the common bile duct was measured on the soft tissue biomechanical testing machine to calculate the compliance. Transection, frozen sections, HE staining, with a computer image analysis system to measure the diameter and wall thickness. Results The diameter of the common bile duct was shorter than that of the adult (P <0.01). The thickness and diameter of common bile duct in 7 ~ 10 months old pigs were similar to those of adults (P> 0.05 ). The compliance of pig bile duct in 3 ~ 6 months and 11,12 months old was significantly less than that in adults (P <0.01). The compliance of bile duct in 7 ~ 10 months old pig was similar to that in adults (P> 0.05). Conclusion The geometry and compliance of the common bile duct in pigs aged 7-10 months are similar to those in adults. From the aspect of biomechanics, it is possible for pig liver from 7 to 10 months old to serve as the donor liver for human xenografts.
