目的:探讨玉女煎合生脉散对围绝经期大鼠去甲肾上腺素(NE)与多巴胺(DA)及5-羟色胺(5-HT)含量影响.方法:选取Wistar雌性大鼠40只,对大鼠连续进行阴道涂片,棉签沾生理盐水后置入大鼠阴道中旋转,获取少量脱落细胞及分泌物,在载玻片内均匀涂抹,而后进行HE染色,共镜检15天,表现为无动情周期可确定是围绝经期大鼠.随机数字表法将大鼠分成四组,模型组(10只)、玉女煎低剂量组(10只)、玉女煎中剂量组(10只)及玉女煎高剂量组(10只);玉女煎低、中、高剂量组大鼠分别灌胃玉女煎合生脉散1.25、2.50、3.75g/kg,模型组大鼠灌胃同剂量生理盐水,每天一次,连续灌胃四周.ELISA法检测血清促卵泡生成激素(FSH)、雌二醇(E2)、促黄体生成激素(LH)、孕酮(P)、DA、5-HP及下丘脑内神经递质β-内啡肽(β-EP)、NE含量,取大鼠脑组织,免疫组化检测下丘脑内促必激素释放激素(GnRH)、雌二醇受体(ER)蛋白表达状况.结果:玉女煎中剂量组、玉女煎高剂量组大鼠血清LH、FSH含量较模型组、玉女煎低剂量组显著降低,玉女煎中剂量组、玉女煎高剂量组大鼠血清E2、P含量较模型组、玉女煎低剂量组显著升高,差异均有统计学意义(P<0.05);玉女煎中剂量组、玉女煎高剂量组大鼠血清β-EP、NE、5-HT及DA含量较模型组、玉女煎剂量组显著升高,差异均有统计学意义(P<0.05);玉女煎中剂量组、玉女煎高剂量组大鼠GnRH表达较模型组、玉女煎低剂量组显著降低,玉女煎中剂量组、玉女煎高剂量组大鼠ER表达较模型组、玉女煎低剂量组显著升高,差异均有统计学意义(P<0.05).结论:玉女煎合生脉散可显著改善围绝经期大鼠血清性激素、单胺类神经递质含量,对围绝经期大鼠内分泌功能紊乱有调节影响.“,”Objective:To analyze the effect of Yu'nv Decoction combined with Shengmai Powder on the levels of norepinephrine (NE), dopamine (DA) and 5-hydroxytryptamine (5-HT) for perimenopausal rats.Methods:40 Wistar female rats were selected and consecutively subjected to vaginal smear.Cotton swabs were immersed in physiological saline and placed in the vagina to rotate.A small amount of exfoliated cells and secretions were obtained, uniformly smeared in slides, and then stained with HE.A total of 15 days of co-microscopic examination showed that there was no estrus cycle and it was confirmed that it was a perimenopausal rat.Rats were divided into four groups by a random number table method.The rats in the model group (10 rats), the Yu'nv Decoction low-dose group (10 rats), the Yu'nv Decoction medium-dose group (10 rats), and the Yu'nv Decoction high-dose group (10 rats).Rats in the low-dose, medium-dose, and high-dose groups were given intragastric administration with 1.25, 2.50, and 3.75 g/kg of Yu'nv Decoction combined with Shengmai Powder respectively.Rats in the model group were given the same dose of normal saline by gavage.All them, once a day for four consecutive weeks.Detection of serum FSH, E2, LH, progesterone (P), DA, 5-HP and hypothalamus neurotransmitter β-endorphin (β-EP) and NE contents were detected by ELISA.Took the rat brain tissue, the expression of GnRH and estradiol receptor (ER) proteins in the hypothalamus was detected by immunohistochemistry.Results:The levels of serum LH and FSH in rats of medium-dose and high-dose group were significantly lower than those of the model group and the low dose group.The contents of serum E2 and P were higher in the medium-dose and high-dose group than those of the model group and the low dose group.The differences were statistically significant (P < 0.05).The levels of serum β-EP, NE, 5-HT, and DA in the medium-dose and the high-dose group were significantly higher than those in the model group and the low dose group, and the differences were statistically significant (P < 0.05).The expression of GnRH in the medium-dose and the high-dose group were significantly lower than that in the model group and the low dose group.The expression levels of ER in rats of the medium-dose group and the high-dose group were significantly higher than that in the model group and the low-dose group, and the differences were statistically significant (P < 0.05).Conclusion:Yu'nv Decoction combined with Shengmai Powder can significantly improve the levels of serum sex hormones and monoamine neurotransmitters in perimenopausal rats, and have a regulatory effect on endocrine dysfunction in perimenopausal rats.
