目的 探讨孕激素对体外培养的人子宫内膜样癌(endometrioid carcinoma, EC)JEC细胞中p57kip2、Cyclin E表达的影响。方法 分别采用高、中、低三种浓度(u0018×u00180-4、u0018×u00180-6、u0018×u00180-8 mol/L)的孕酮(progesterone, P)处理体外培养的JEC细胞,以未加药JEC细胞作为对照组,作用24、48、72 h后,分别采用光学显微镜和电子显微镜观察JEC细胞的形态变化,MTT法检测JEC细胞的生长情况,Western blot法检测JEC细胞中p57kip2、Cyclin E蛋白的表达。结果 MTT法、光学显微镜、电子显微镜结果显示,中、低浓度P可促进JEC细胞的生长(P<0.05),高浓度P则抑制JEC细胞的生长(P<0.05);Western blot结果显示,P作用于JEC细胞后:(u0018)作用48、72 h时,随着P浓度的增加,p57kip2蛋白的表达逐渐增高(P>0.05),Cyclin E蛋白的表达则逐渐降低(P<0.05);(2)同一浓度时,随着P作用时间的延长,p57kip2蛋白在高、中、低浓度组中的表达均呈递增趋势(P<0.05),Cyclin E蛋白在中、高浓度组的表达均呈递减趋势(P<0.05);p57kip2和Cyclin E蛋白的表达呈负线性相关关系(P<0.05)。结论 在EC的发生发展中,孕酮对p57kip2、Cyclin E蛋白的表达均具有一定的调控作用。
Objective To investigate the effects of progesterone on the expression of p57kip2 and Cyclin E in human endometrioid carcinoma (EC) JEC cells cultured in vitro. Methods Progesterone (P) with high, medium and low concentrations (u0018 × u00180-4, u0018 × u00180-6, u0018 × u00180-8 mol / L) JEC cells were treated with unadministered JEC cells for 24, 48 and 72 hours. The morphological changes of JEC cells were observed by optical microscope and electron microscope respectively. The growth of JEC cells was detected by MTT assay and Western blot Expression of p57kip2 and Cyclin E in JEC cells. Results The results of MTT, light microscope and electron microscope showed that medium and low concentrations of P could promote the growth of JEC cells (P <0.05), and high concentrations of P inhibited the growth of JEC cells (P <0.05). Western blot results showed that P After treated with JEC cells for 48 and 72 h, the expression of p57kip2 protein gradually increased (P & gt; 0.05) and the expression of Cyclin E protein decreased gradually (P & lt; 0.05) with the increase of P concentration. (2) At the same concentration, the expression of p57kip2 protein in the high, medium and low concentration groups showed an increasing trend with the prolongation of P (P <0.05). The expression of Cyclin E protein in medium and high concentration groups (P <0.05). There was a negative linear correlation between the expression of p57kip2 and Cyclin E protein (P <0.05). Conclusions Progesterone regulates the expression of p57kip2 and Cyclin E in the pathogenesis of EC.
