【目的】优化能源作物甜高粱组培苗的生根培养体系,为遗传转化奠定基础。【方法】利用不同基本培养基(MS、1/2MS)对组培苗生根的影响进行筛选试验,得到较有利于生根的基本培养基;在最佳培养基中添加不同浓度的植物激素(IBA、NAA、ABT),综合根的质量、生根率、生根时间、生根苗的长势等筛选最佳生根培养基配方。【结果】筛选出最佳生根培养基组合:北甜三号(BT3)为1/2MS+2.0mg/LIBA,辽甜三号(LT3)为1/2MS+3.0mg/LIBA,且根粗、苗壮,两品种的生根率存在差异。此外,未加激素的MS和1/2MS培养基均无组培苗生根。【结论】在1/2MS上添加2.0mg/LIBA或3.0mg/LIBA分别为北甜三号、辽甜三号组培苗生根的最佳途径。
【Objective】 Optimize the rooting culture system of tissue culture seedlings of sweet sorghum, and lay the foundation for genetic transformation. 【Method】 The effects of different basic medium (MS, 1 / 2MS) on the rooting of tissue culture seedlings were screened to obtain the basic medium more conducive to rooting. Different concentrations of plant hormone (IBA , NAA, ABT), root quality, rooting rate, rooting time, the growth of rooting seedlings and so on. 【Result】 The best combination of rooting medium was screened out: BT3 was 1 / 2MS + 2.0mg / LIBA, LT3 was 1 / 2MS + 3.0mg / LIBA, Miao Zhuang, rooting rate of the two varieties are different. In addition, no hormone-free MS and 1 / 2MS medium without rooting tissue culture seedlings. 【Conclusion】 The best way to rooting of tissue culture seedlings of Beitang No. 3 and Liao Tian No. 3 with 2.0 mg / L IBA or 3.0 mg / L IBA on 1/2 MS, respectively.
