FK506(FK)是日本Fujisawa公司的Goto等于1984年从筑波山土壤中链霉菌属(Streptomycstrukufaensiz)放线菌中分离出来的发酵产物,化学结构属23元环大环内酯类抗生素,分子量822,易溶于甲醇、乙醇、丙酮,不溶于水。FK是目前已知最强的免疫抑制剂,其效价较CsA(Cyclosporine A)强100倍以上,经5年动物试验,1989年已开始临床使用,因其作用强,副作用较CsA少,正在取代CsA作为移植术后免疫抑制剂首选药的地位。
