为了构建能同时发酵葡萄糖和木糖的酵母重组菌株,采用双灭活原生质体技术,通过聚乙二醇将树干毕赤酵母(Pichia stipitis)NRRL Y-7124与酿酒酵母(Saccharomyces cerevisiae)NRRL Y-2034进行融合,选育出一株遗传性能稳定且发酵木糖产乙醇性能显著提高的优良融合菌株F11.融合子F11在60g/L木糖中发酵,乙醇产量较亲本提高30.27%,达到20.09g/L;在60g/L混合糖(葡萄糖∶木糖=1∶3)发酵中,融合子F11的乙醇最高产量为22.52g/L,比亲本提高29.7%,同时融合子的耐乙醇能力也明显增强.
