目的探讨核因子NF-κBP50、NF-κBP65在骨质疏松发病过程中的意义。方法选用4个月龄雌性BALB/C小鼠随机分假手术对照组(CON组)和去卵巢骨质疏松模型组(OVX组),术后第12周测量全身骨密度后处死,取股骨下端用ABC免疫组织化学、原位杂交方法测定骨组织中核因子的蛋白含量及表达水平并进行细胞定位。结果OVX组与CON组相比,骨密度下降,股骨骨小梁稀疏、成骨细胞数目减少而破骨细胞数目增多;免疫组织化学染色结果显示,NF-κBP50、NF-κBP65蛋白在模型组和对照组均有阳性表达,主要位于骨小梁周边成骨细胞、骨细胞及骨髓细胞胞浆中,OVX组蛋白含量均高于CON组,且与骨密度呈负相关(P<0·01或P<0·05);原位杂交结果表明,OVX组NF-κBP50mRNA、NF-κBP65mRNA表达显著高于CON组(P<0·05)。结论骨质疏松骨组织及骨髓中核因子的蛋白含量及表达水平升高,在骨质疏松骨微环境中的表达异常并与骨密度明显相关。
Objective To investigate the significance of NF-κBP50 and NF-κBP65 in the pathogenesis of osteoporosis. Methods 4-month-old female BALB / C mice were randomly divided into sham-operated control group (CON group) and ovariectomized osteoporosis model group (OVX group). Whole body BMD was measured after 12 weeks of operation. ABC immunohistochemistry and in situ hybridization were used to determine the protein content and expression level of nuclear factor in bone tissue and cell location. Results Compared with CON group, the bone mineral density decreased, the trabecular bone became thinner, the number of osteoblasts decreased and the number of osteoclasts increased in OVX group. Immunohistochemical staining showed that the expressions of NF-κBP50 and NF-κBP65 in model group and The expression of OVX protein in OVX group was higher than that in CON group, and negatively correlated with bone mineral density (P <0.01 or P <0.05). In situ hybridization results showed that the expression of NF-κBP50mRNA and NF-κBp65mRNA in OVX group was significantly higher than that in CON group (P <0.05). Conclusion The protein content and expression of osteonectin in bone tissue and bone marrow of osteoporosis are abnormal, and are abnormally expressed in the bone microenvironment of osteoporosis and are significantly correlated with bone mineral density.
