采用凝胶渗透色谱法检测反应温度、反应时间和酶液饱和度对裂褶多糖在酶解过程中的分子量及分子量分布的影响。结果表明,内切β-1,3-葡聚糖酶降解裂褶多糖可采用饱和度80%的酶液,最适反应温度为50℃,反应时间控制在2 h以内能得到较好的降解效果。利用凝胶渗透色谱法可以有效监控裂褶多糖降解反应程度。
The effects of reaction temperature, reaction time and enzyme liquid saturation on the molecular weight and molecular weight distribution of Schizophyllan polysaccharide during enzymolysis were investigated by gel permeation chromatography. The results showed that the degradation rate of endogenous β-1,3-glucanase polysaccharide can be used 80% saturation of the enzyme solution, the optimum reaction temperature was 50 ℃, the reaction time controlled within 2 h to get better degradation effect. Gel permeation chromatography can effectively monitor the degree of degradation of polysaccharide from Schizophyllan.
