目的如何对小鼠动脉粥样硬化斑块体积进行定量分析国内尚未见报到。本文将以载脂蛋白E基因敲除小鼠为例介绍一种确定小鼠动脉粥样硬化斑块体积的新方法。方法将16周龄雌性ApoE基因敲除小鼠的主动脉根部进行连续冰冻切片700μm,每间隔100μm收集组织切片进行油红O染色,利用Leica Q500MC图像分析软件测量斑块面积,利用Cavalieri公式计算动脉粥样硬化斑块体积。结果本例中小鼠主动脉根部粥样斑块面积为520 632μm2,体积为364 442μm3。结论本文介绍的确定小鼠动脉粥样硬化斑块体积的方法操作简单、易于进行标准化。
Objective How to quantify the size of atherosclerotic plaque in mice has not been reported in China. In this paper, apolipoprotein E knockout mice will be taken as an example to introduce a new method to determine the size of atherosclerotic plaque in mice. Methods The aorta of 16-week-old female ApoE knockout mice were serially frozen sectioned at 700μm. The tissue sections were collected at intervals of 100μm for oil red O staining. The area of plaque was measured by Leica Q500MC image analysis software. The artery was calculated by Cavalieri formula Atherosclerotic plaque volume. Results In this case, the aortic root atherosclerotic plaque area was 520 632 μm 2 and the volume was 364 442 μm 3. Conclusion The method described in this paper to determine the volume of atherosclerotic plaque in mice is simple and easy to standardize.
