大白鼠主要组织相容性复合体(RT1-B)编码两套Ⅱ类分子(Ia抗原)称为A和E,每个Ⅱ类分子包括两条糖蛋白链称为Aα和Aβ或Eα和Eβ。本文作者使用mRNA选择实验(mRNA selectionassay),由大白鼠脾细胞富含编码大白鼠Aα链mRNA的poly(A)mRNA制得一个cDNA文库。从开始的192个克隆筛选中,鉴定出一个特异地选择大白鼠Aα链mRNA的cDNA克隆,定名为pRIa1,部分限制性内切酶分析证明此cDNA以大约600bp长度插入到pRIa.1中,且只有一个Pst1切点,从pRIa.1中取出EcoR1-Pst1切点之间的150个bp作为杂交探针从整个脾poly(A)mRNA制得的cDNA中
