目的:建立大豆黄卷总黄酮的测定方法,同时对其水煎煮过程中的提取动力学过程进行模拟,建立动力学方程并验证。方法:采用紫外分光光度法,筛选对照品和测定波长,并对其方法学进行考察;使用该方法,测定不同提取时间和溶剂下大豆黄卷水提浓度,以简化的数学动力学模型为基础,推算出方程参数,将测定结果代入进行方程拟合。结果:染料木苷在0.0009296~0.01162 g·L-1具有良好的线性关系,回归方程为Y=54.859X-0.0034,r=0.9999,平均回收率为103.6%,RSD=0.83%;拟合的动力学模型结果为CB=(0.1520×t0.5/M-1.7970)0.5807,验证结果良好。结论:所建立的大豆黄卷总黄酮测定方法稳定可行,重现性好。模型适用并能良好的描述大豆黄卷水提取的动力学过程。
OBJECTIVE: To establish a method for the determination of total flavonoids in soybean yellow curd, and to simulate the extraction kinetics during the decoction of water, establish the kinetic equation and verify. Methods: UV spectrophotometry was used to screen the reference substance and determine the wavelength. The method was used to determine the concentration of soybeans in different extraction time and solvent. Based on the simplified mathematical model , Calculate the equation parameters, the measured results into the equation fitting. Results: The genistein showed good linearity in the range of 0.0009296 ~ 0.01162 g · L-1, the regression equation was Y = 54.859X-0.0034, r = 0.9999, the average recovery was 103.6% and the RSD was 0.83% The result of the model is CB = (0.1520 × t0.5 / M-1.7970) 0.5807, and the verification result is good. Conclusion: The method of determination of total flavonoids in soybean volume is stable and feasible with good reproducibility. The model is suitable and can well describe the kinetic process of soybean yellow roll water extraction.
