目的探讨犀角地黄汤对内毒素诱导下持续性大脑中动脉栓塞模型(pMCAO)大鼠脑组织的自噬相关LC3-II蛋白的表达;以及透射电镜下观察双层膜的自噬体。方法造模后进行分组,并采用不同剂量犀角地黄汤进行干预,分别在透射电镜下观察自噬体;检测自噬体膜标志性蛋白质LC3-II的表达。结果模型组有明显的空泡现象出现,中等剂量的犀角地黄汤治疗组有明显空泡现象。造模后第3天和第6天,LC3-II蛋白表达增强,LC3-II/LC3-l比值增加(P<0.05)。结论细胞质LC3-I脂质化形成LC3-Ⅱ。中高剂量组明显降低LC3-II蛋白含量,中剂量组效果更佳,能更好地抑制自噬相关蛋白的表达。
Objective To investigate the expression of LC3-II protein in the brain of rats with endotoxin-induced persistent middle cerebral artery occlusion (pMCAO) induced by Xijiao Rehmanniae Decoction and to observe the autophagosome of bilayer membrane under transmission electron microscope. Methods After modeling, the rats were divided into groups according to rhizoma dioscoreae decoction. The autophagosomes were observed under transmission electron microscope. The expression of LC3-II, a marker protein of autophagosome, was detected. Results In the model group, obvious vacuoles appeared and the medium-dose rhino-horn-rhubarb decoction group had obvious vacuoles. LC3-II protein expression was increased and LC3-II / LC3-l ratio increased on the 3rd and 6th day after model establishment (P <0.05). Conclusion Cytoplasmic LC3-I lipids form LC3-II. The medium and high dose groups significantly reduced the content of LC3-II protein, and the middle dose group had better effect, which could better inhibit the expression of autophagy-related proteins.
