目的探讨结肠癌转移相关基因1(MACC1)和肝细胞生长因子受体蛋白(c-Met)表达与宫颈癌盆腔淋巴结转移相关性。方法选取宫颈癌患者113例,宫颈上皮内瘤变Ⅱ~Ⅲ患者41例,正常宫颈组织患者47例,利用免疫组化SP法检测MACC1和c-Met在组织中表达,利用二分类Logistic回归分析对宫颈癌盆腔转移相关因素进行分析。结果 MACC1和c-Met在宫颈癌组织中相对表达量分别为(7.64±1.37)和(6.73±1.22),均高于宫颈上皮内瘤变Ⅱ~Ⅲ组织和正常宫颈组织,且宫颈上皮内瘤变Ⅱ~Ⅲ组织高于正常宫颈组织,差异均有统计学意义(P<0.05);Spearman相关分析显示,MACC1在宫颈癌组织中表达与c-Met呈正相关(r=0.467,P<0.05);宫颈癌发生盆腔淋巴结转移与脉管浸润、肌层浸润深度、MACC1阳性表达、cMet阳性表达有关(P<0.05),多因素Logistic回归分析显示,肌层浸润深度、MACC1阳性表达和c-Met阳性表达是宫颈癌发生盆腔淋巴结转移的独立危险因素(P<0.05)。结论 MACC1和c-Met在宫颈癌组织中呈高表达,可能共同参与了宫颈癌发生及进展,是宫颈癌发生盆腔淋巴结转移的独立危险因素。
Objective To investigate the correlation between the expression of metastasis-associated gene 1 (MACC1) and hepatocyte growth factor receptor (c-Met) in colon cancer and pelvic lymph node metastasis of cervical carcinoma. Methods 113 cases of cervical cancer, 41 cases of cervical intraepithelial neoplasia Ⅱ ~ Ⅲ patients and 47 cases of normal cervical tissue were selected. The expression of MACC1 and c-Met was detected by immunohistochemical SP method. The factors related to pelvic metastasis of cervical cancer were analyzed. Results The relative expression levels of MACC1 and c-Met in cervical cancer tissues were (7.64 ± 1.37) and (6.73 ± 1.22), respectively, which were higher than those in cervical intraepithelial neoplasia tissues Ⅱ ~ Ⅲ and normal cervical tissues. Cervical intraepithelial neoplasia The results of Spearman correlation analysis showed that there was a positive correlation between the expression of MACC1 and c-Met in cervical cancer (r = 0.467, P <0.05), and the difference was statistically significant . The pelvic lymph node metastasis of cervical carcinoma was related to the vascular infiltration, the depth of myometrial invasion, the expression of MACC1 and the expression of cMet (P <0.05). Logistic regression analysis showed that the depth of myometrial invasion, the expression of MACC1 and the expression of c-Met The positive expression was an independent risk factor of pelvic lymph node metastasis in cervical cancer (P <0.05). Conclusions MACC1 and c-Met are highly expressed in cervical cancer and may participate in the occurrence and progression of cervical cancer. It is an independent risk factor of pelvic lymph node metastasis in cervical cancer.
