[目的]提取并测定长白山区野生菟丝子(Guscuta japonica Choisy)中多糖的含量。[方法]采用超声技术提取野生菟丝子中的多糖,用苯酚-硫酸法测定多糖含量,以葡萄糖(105℃干燥至恒重)为对照品。[结果]在9.440~66.080μg/ml内,样品的浓度与吸光度呈良好的线性关系,线性回归方程为:A=0.0096C+0.0033(r=0.9999),加标回收率为99.95%(n=5),RSD=0.07%,长白山区野生菟丝子中多糖平均含量为5.31%。[结论]长白山区野生菟丝子中多糖含量较高,具有较高的药用价值和开发前景。
[Objective] The research aimed to extract and determine the polysaccharide content of Guscuta japonica Choisy in Changbai Mountains. [Method] The polysaccharide in wild dodder was extracted by ultrasonic technique. The content of polysaccharide was determined by phenol - sulfuric acid method. The reference substance was dextrose (105 ℃ dried to constant weight). [Results] The linearity of the sample was linear with the absorbance in the range of 9.440 ~ 66.080μg / ml. The linear regression equation was A = 0.0096C + 0.0033 (r = 0.9999) and the recovery was 99.95% 5), RSD = 0.07%, the average content of polysaccharide in wild dodder was Changbai Mountain was 5.31%. [Conclusion] The content of polysaccharide in wild dodder was high in Changbai Mountain, which had high medicinal value and development prospect.
