目前食用菌菌种的保藏材料都取用其菌丝体。保藏方法大致有三种:一为菌丝体的继代培养;二为菌丝体的液体石蜡保藏;三为菌丝体的液氮低温保藏。继代培养法使用方便,但传代次数多了,菌种易衰老退化;液体石蜡和液氮低温保藏法,保藏时间长,但不易寄运。用液体石蜡保藏的菌种也易遭霉菌污染;用液氮低温法保藏又需特殊的液氮冰箱。本试验则以食用菌的担孢子为保藏材料,以滤纸条为保藏基质,即让食用菌孢子保藏在滤纸条上,并密封于试管内。此法保藏时间长,制备简便,又易贮运。
At present, the preservative material of edible fungi takes its mycelium. There are three kinds of preservation methods: one is subculture of mycelium; the other is liquid paraffin of mycelium; the other is mycelium liquid nitrogen cryopreservation. Subculture method is easy to use, but the passage number of times, the strain is easy to aging degradation; liquid paraffin and liquid nitrogen cryogenic preservation method, the preservation of a long time, but not easy to send. Stored with liquid paraffin bacteria are also susceptible to mold contamination; liquid nitrogen cryogenic preservation method also need a special liquid nitrogen refrigerator. In this experiment, basidiospores of edible fungi are used as the preservation material, and the filter paper is used as the preservation matrix, that is, the edible fungus spores are deposited on the filter paper and sealed in a test tube. This method is preserved for a long time, preparation is simple, easy storage and transportation.
