背景:应用抖笼换能器法、阈上、阈下剂量戊巴比妥钠睡眠时间的实验法,探讨橙皮水提取物对昆明种小鼠睡眠和自发活动的影响。目的:观察橙皮水提取物对昆明种小鼠睡眠和自发活动的影响。设计:随机对照实验观察。单位:赣南医学院病理教研室、生理学教研室及药理教研室。材料:实验于2005-01/02在赣南医学院科研中心实验室完成。选择健康成年昆明种清洁级小鼠90只,每种实验方法30只,随机分为3组,对照组,小剂量给药组,大剂量给药组;每组10只。方法:①采用抖笼换能器法记录小鼠自发活动情况,小波(波幅<5mm)为小鼠轻度活动(舐毛、瘙痒)的波形;中波(波幅5~10mm)为小鼠中度活动(走动、立起、爬上、爬下)的波形;大波(波幅>10mm以上)为小鼠强度活动(跑动、上串下跳)的波形。小剂量给药组、大剂量给药组分别腹腔注射橙皮水提取物5mg/g、10mg/g和对照组注射等量生理盐水,15min后放回吊笼中,稳定3min后记录15s内小鼠活动波形。②橙皮水提取物对小鼠阈上或阈下剂量戊巴比妥钠睡眠时间的实验法:小剂量给药组、大剂量给药组分别腹腔注射橙皮水提取物5mg/g、10mg/g,对照组注射等量生理盐水;15min后,3组均腹腔注射阈上剂量的戊巴比妥钠(2.5g/L)0.02mL/g,或阈下剂量的戊巴比妥钠(2.5g/L)0.01mL/g,比较给药组与对照组动物的入睡个数、入睡时间和睡眠持续时间。主要观察指标:①小鼠自发活动次数。②阈上剂量戊巴比妥钠小鼠睡眠时间。③阈下剂量戊巴比妥钠小鼠睡眠时间。结果:各实验组小鼠无脱失情况,全部进入结果分析。①橙皮水提取物小剂量给药组,大剂量给药组使中波、大波出现次数显著减少,对小鼠自发活动具有明显的抑制作用,[(90.5±14.7),(58.3±12.5),(45.6±10.1),t=2.341~3.215;P<0.05或P<0.01]。②橙皮水提取物小剂量给药组,大剂量给药组能显著加速阈上剂量戊巴比妥钠的入睡时间和延长戊巴比妥钠的睡眠时间,差异有显著性意义。③橙皮水提取物小剂量给药组,大剂量给药组能显著加速阈下剂量戊巴比妥钠的入睡时间和延长戊巴比妥钠的睡眠时间,差异有非常显著性意义。结论:橙皮水提取物具有明显的延长昆明种小鼠睡眠时间和抑制小鼠自发性活动。与戊巴比妥钠有协同的中枢抑制作用。
BACKGROUND: The method of shaking cage transducer, suprathreshold and subthreshold doses of sodium pentobarbital for sleeping time was used to study the effects of orange peel extract on the sleep and spontaneous activities of Kunming mice. Objective: To observe the effect of orange peel extract on sleep and spontaneous activity in Kunming mice. Design: Randomized controlled experimental observations. Unit: Department of Pathology, Department of Physiology, and Department of Pharmacology, Wannan Medical College. MATERIALS: The experiment was performed at the Laboratory of Scientific Research Center of Wannan Medical College from January to February 2005. A total of 90 healthy Kunming species of clean grade mice were selected, 30 of each experimental method were randomly divided into 3 groups, control group, low dose group, and large dose group; 10 in each group. Methods: 1 The voluntary cage transducer method was used to record the spontaneous activity of mice. The wavelet (wave amplitude <5mm) was the waveform of mild activity (broiler hair and pruritus) in mice. Medium wave (wave amplitude 5~10mm) was used in mice. The waveform of the activity (walking, erecting, climbing, climbing down); the large wave (wave amplitude >10mm) is the waveform of the intensity activity (running, jumping up and down) of the mouse. In the low-dose group and large-dose group, 5 mg/g orange peel extract and 10 mg/g saline were injected intraperitoneally, and the same amount of saline was injected into the control group. After 15 min, the saline was put back into the cage, and the stability was recorded within 15s after being stabilized for 3 min. Mouse activity waveform. 2Experimental study on the time of pentobarbital sodium sleeping in the suprathreshold or subthreshold doses of orange peel extract in mice: Intraperitoneal injection of orange peel extract 5 mg/g, 10 mg in the low-dose group and large-dose group /g, The control group was injected with equal volume of normal saline; after 15 minutes, all three groups received an intraperitoneal injection of a suprathreshold dose of sodium pentobarbital (2.5 g/L) of 0.02 mL/g, or a subliminal dose of pentobarbital sodium ( 2.5g/L) 0.01mL/g, comparing the number of sleeping, sleeping time and sleep duration between the administration group and the control group. MAIN OUTCOME MEASURES: 1 Number of spontaneous activity in mice. 2 threshold dose of sodium pentobarbital in mice sleep time. 3 subthreshold dose of sodium pentobarbital in mice sleep time. RESULTS: There was no loss of mice in all experimental groups, and all of them entered the analysis of results. 1 The orange peel extract was administered in small dose group, the large dose group significantly reduced the occurrence of medium and large waves, and had a significant inhibitory effect on spontaneous activity of mice [(90.5±14.7), (58.3±12.5) (45.6±10.1), t=2.341 to 3.215; P<0.05 or P<0.01]. 2 In the small dose group of orange peel extract, the large dose group could significantly accelerate the sleep time of the suprathreshold dose of pentobarbital sodium and prolong the sleep time of pentobarbital sodium, and the difference was significant. 3 In the small dose group of orange peel extract, the large dose group could significantly accelerate the sleep time of the subthreshold dose of pentobarbital sodium and prolong the sleep time of pentobarbital sodium, and the difference was very significant. Conclusion: The orange peel extract has obvious prolongation of sleep time and inhibits spontaneous activity in mice. Synergistic central inhibition with pentobarbital sodium.
