目的探讨乳杆菌对小肠上皮钙离子吸收的影响及其作用机制。方法测定植物乳杆菌ST III发酵上清液对复合钙盐解离效果的基础上,通过MTT比色法测定植物乳杆菌ST III发酵上清液对大鼠小肠隐窝上皮细胞(IEC-6)增殖率影响以及用fluo-3AM荧光染色观察对IEC-6细胞内游离钙离子浓度变化。结果植物乳杆菌ST III发酵上清液对大鼠IEC-6增殖有明显的促进作用,与对照组比较差异有统计学意义,24h(F=548.401,P=0.000,P<0.05),48h(F=722.352,P=0.000,P<0.05),并呈良好的量效关系;同时植物乳酸菌ST III发酵上清液刺激后小肠隐窝上皮细胞6、12和24h,细胞内游离钙离子荧光强度相对值明显高于对照组,6h(F=203.911,P=0.000,P<0.05),12h(F=106.158,P=0.000,P<0.05),24h(F=26.076,P=0.000,P<0.05),差异有统计学意义。结论植物乳杆菌ST III在细胞水平上促进小肠上皮增殖并对细胞内Ca2+有明显的促进作用。
Objective To investigate the effect of Lactobacillus on calcium absorption in intestinal epithelium and its mechanism. Methods Based on the dissociation effect of Lactobacillus plantarum ST III fermentation supernatant on composite calcium salt, the effects of Lactobacillus plantarum ST III fermentation supernatant on the intestinal crypt epithelial cells (IEC-6) Proliferation rate and the change of intracellular free calcium concentration in IEC-6 cells by fluo-3AM fluorescence staining. Results Lactobacillus plantarum ST III fermentation supernatant significantly promoted the proliferation of IEC-6 in rats, which was significantly different from the control group (P <0.05) F = 722.352, P = 0.000, P <0.05), and showed a good dose-effect relationship. Meanwhile, the intestinal crypt epithelial cells stimulated by Lactobacillus plantarum ST III fermentation supernatant for 6, 12 and 24 h, the intracellular free calcium fluorescence intensity The relative value was significantly higher than that of the control group at 6h (F = 203.911, P = 0.000, P <0.05), 12h (F = 106.158, P = 0.000, P < 0.05), the difference was statistically significant. Conclusion Lactobacillus plantarum ST III can promote intestinal epithelial proliferation and promote intracellular Ca2 + at the cellular level.
