目的探讨肺癌组织中线粒体转录因子A的表达与线粒体DNA拷贝数变化的关系。方法采用实时荧光定量PCR和RT-PCR方法,对肺癌及相应正常肺组织中mtTFA mRNA的表达及其与线粒体DNA拷贝数变化的关系进行了检测和分析。结果癌组织和癌旁正常组织mtTFA mRNA的表达量之间无明显差异(P=0.221)。线性回归分析结果表明,癌旁正常组织中,mtTFA mRNA的表达量和线粒体DNA拷贝数之间有直接的相关性(r=0.910,P<0.01);而在癌组织中,线粒体DNA拷贝数和mtTFA mRNA的表达水平无关(r=0.135,P>0.05)。结论 mtDNA转录、复制的核调节因子mtTFA的表达水平和肺癌线粒体DNA拷贝数之间无相关性。
Objective To investigate the relationship between mitochondrial DNA transcription factor A and mitochondrial DNA copy number in lung cancer. Methods The expression of mtTFA mRNA and its relationship with mitochondrial DNA copy number in lung cancer and corresponding normal lung tissues were detected by real-time fluorescence quantitative PCR and RT-PCR. Results There was no significant difference in the expression of mtTFA mRNA between adjacent tissues and normal tissues (P = 0.221). The results of linear regression analysis showed that there was a direct correlation between mtTFA mRNA expression and mitochondrial DNA copy number (r = 0.910, P <0.01), while in normal tissues, mitochondrial DNA copy number and mtTFA mRNA expression was not related (r = 0.135, P> 0.05). Conclusion There is no correlation between the mtTFA expression level in mtDNA transcription and replication and mitochondrial DNA copy number in lung cancer.
