目的初步建立预发酵-双相联合酸水解法从黄姜药材中萃取薯蓣皂素的工艺。方法采用正交实验设计法,以薯蓣皂素的得率为考察指标,分别优化预发酵和双相联合酸水解法的最佳条件,并与两类传统提取方法进行比较。结果将黄姜药材粉末在500 r·min-1搅拌下于30℃预发酵72 h后,加入浓硫酸和甲醇使它们的终浓度均为10%,再加入石油醚形成双相体系,并于120℃油浴中加热回流萃取4 h,薯蓣皂素的平均得率为4.28%;而两类传统水解方法的得率则分别为3.01%和3.28%。结论利用预发酵-双相联合酸水解法从黄姜药材中萃取薯蓣皂素简便易行,并且提取率明显高于其他方法。
Objective To establish a pre-fermentation-biphasic acid hydrolysis process of diosgenin extraction from turmeric. Methods The optimal conditions of pre-fermentation and biphasic combined acid hydrolysis were optimized by orthogonal experimental design with the yield of diosgenin as index. The two methods were compared with two traditional extraction methods. Results After the yellow ginger medicinal powder was pre-fermented at 500 r · min-1 for 72 h at 30 ℃, concentrated sulfuric acid and methanol were added to make their final concentration of 10%, then petroleum ether was added to form a biphasic system. ℃ oil bath heating reflux extraction 4 h, the average yield of diosgenin was 4.28%; while the yield of the two types of traditional hydrolysis methods were 3.01% and 3.28% respectively. Conclusions The extraction of diosgenin from turmeric with pre-fermentation-biphasic acid hydrolysis is simple and convenient, and the extraction rate is obviously higher than other methods.
