作者从1例急性单核细胞白血病男性患儿的周围血培育出一株单核细胞白血病细胞系(THP-1)。培养液为 RPMI-1640加20%胎牛血清。在带 Molton 塞的试管含5%CO_2的二氧化碳培养箱中,患儿的白细胞一经培养即可很快繁殖,六个月后细胞适应于塞普通塞子的培养瓶。THP-1细胞在悬浮培养中呈松散的团块,不贴壁。倍增时为60~70小时。培养细胞的形态与培养前相似。胞体圆形或稍呈多边形,直径12~14微米,胞浆量中等嗜碱性,含有小的嗜天青颗粒和几个空泡。核有凹陷,形状不规则。透射电镜观察见核有深折,核
The authors generated a monocytic leukemia cell line (THP-1) from the peripheral blood of 1 male patient with acute monocytic leukemia. The culture medium was RPMI-1640 plus 20% fetal calf serum. In a 5% CO 2 CO2 incubator with a Molton plug, the children’s white blood cells multiply rapidly after culturing, and six months later, the cells adapt to the culture plug of a regular plug.
