目的:体外观察全反式维甲酸(ATRA)对卵黄囊瘤细胞系细胞体外生长的影响。方法:卵黄囊瘤细胞系细胞持续培养在含ATRA的96孔板中,采用MTT法检测ATRA的体外抗增殖效果,采用流式细胞仪检测ATRA对细胞周期的影响,用相差倒置显微镜及透射电镜观察细胞形态变化。结果:MTT法检测发现,ATRA作用后细胞的增殖受到了抑制,随着浓度和时间的增加,其抑制作用增强,与对照组比较差异有统计学意义,P<0.05。流式细胞仪检测结果显示,经ATRA处理后的卵黄囊瘤细胞明显阻滞于G1期,S期比例减少,与对照组比较差异有统计学意义,P<0.05。光镜和透射电镜可观察到经ATRA处理后的卵黄囊瘤细胞部分发生形态学的良性分化:超微下见胞核体积缩小且形态规则,核仁减少和消失,细胞表面微绒毛及树枝状伪足消失。对照组细胞无此表现。结论:ATRA对卵黄囊瘤细胞有诱导分化作用,有效抑制了肿瘤细胞的增殖,其机制可能与引起细胞周期阻滞于G1期有关。
Objective: To observe the effect of all-trans retinoic acid (ATRA) on the growth of yolk sac cell line in vitro. Methods: The yolk sac tumor cell lines were cultured continuously in ATRA-containing 96-well plates. The anti-proliferative effect of ATRA in vitro was detected by MTT assay. The effect of ATRA on the cell cycle was detected by flow cytometry. The inverted phase contrast microscope and transmission electron microscopy Observation of cell morphological changes. Results: MTT assay showed that the proliferation of ATRA cells was inhibited after ATRA treatment. The inhibitory effect was enhanced with the increase of concentration and time, and the difference was statistically significant (P <0.05). Flow cytometry results showed that the yolk sac cells treated with ATRA were significantly arrested in G1 phase and decreased in S phase, which was significantly different from the control group (P <0.05). Morphological benign differentiation of yolk sac cells treated with ATRA was observed by light and transmission electron microscopy. The nuclei were reduced in size and morphologically, the nucleoli were reduced and disappeared, and the surface microvilli and dendrites Pseudopods disappear. Control group cells without this performance. CONCLUSION: ATRA induces differentiation of yolk sac cells and effectively inhibits the proliferation of tumor cells. The mechanism may be related to the induction of cell cycle arrest in G1 phase.
