目的:研究不同亚型HPV持续感染的宫颈组织中hTERC基因的表达,并讨论hTERC基因作为诊断不同级别宫颈上皮内瘤变标志物的临床意义。方法:采用荧光原位杂交(fluorescent in situ hybridization,FISH)方法,检测20例HPV转阴宫颈组织及70例持续性HPV感染宫颈组织中hTERC基因的表达;并将hTERC基因的扩增情况与组织病理学诊断结果进行比较。结果:hTERC基因在HPV转阴宫颈组织的阳性率为5.00%;在A5/A6、A7、A9及混合感染组的阳性率分别为:33.33%、53.33%、85.00%及50.00%;A9组hTERC基因扩增与对照组、A5/A6组、A7组及混合感染组比较有统计学差异(P<0.05)。hTERC基因在正常宫颈组织或炎症组、CIN1组、CIN2组及CIN3组的阳性率分别为13.79%、33.33%、66.67%及93.75%,hTERC基因的扩增与宫颈病变的程度呈正相关,hTERC基因在正常宫颈组织或炎症及CIN1组中的扩增与CIN2组、CIN3组比较有统计学差异(P<0.05)。结论:hTERC基因的扩增与宫颈感染HPV的亚型相关,hTERC基因的扩增与宫颈病变程度相关。
OBJECTIVE: To study the expression of hTERC gene in cervical tissues with persistent infection of different subtypes of HPV, and to discuss the clinical significance of hTERC gene as a diagnostic marker for cervical intraepithelial neoplasia of different grades. Methods: The expression of hTERC gene was detected by fluorescent in situ hybridization (FISH) in 20 cases of cervical tissue with cervical lymph node metastasis and 70 cases of persistent HPV infection. The amplification of hTERC gene was compared with that of tissue Pathological diagnosis results were compared. Results: The positive rate of hTERC gene was 5.00% in cervical metastasis of HPV. The positive rate of hTERC gene was 33.33%, 53.33%, 85.00% and 50.00% respectively in A5 / A6, A7, A9 and mixed infection groups. Conclusion: The amplification of hTERC gene is associated with HPV subtype of cervical infection, and the amplification of hTERC gene is associated with the degree of cervical lesions.
