目的观察山稔子油敷剂对深Ⅱ度烧伤小鼠的疗效及其对肿瘤坏死因子-α(TNF-α)、细胞间粘附因子1(ICAM-1)的影响。方法将30只小鼠随机分成正常组,模型组和山稔子油敷剂组,正常组6只,其余两组各12只。造模4 h后给药干预,观察烧伤的一般指标。小鼠分两批处死,4 d时,模型组和山稔子油敷剂组随机处死6只,12 d后再处死余下的小鼠,眼球采血,分离烧伤组织,HE染色观察皮肤组织的病理变化,酶联免疫(Elisa)、免疫组化(IHC)检测烧伤组织TNF-α,ICAM-1的表达。结果 4 d和12 d山稔子油敷剂组创面积明显小于相对应模型组的面积(P<0.05,P<0.01),炎症细胞浸润减少,12 d个别小鼠创面全部愈合。4 d和12 d时山稔子油敷剂组ICAM-1表达均低于模型组(P<0.05),4 d时山稔子油敷剂组TNF-α表达低于模型组(P<0.05),12 d时山稔子油敷剂组明显高于模型组(P<0.05)。结论山稔子油敷剂具有抗炎、减少疤痕、缩短愈合时间的作用,急性期减少TNF-α,修复期维持其相对量的表达量。急性期、修复期均减少ICAM-1的表达。山稔子油敷剂对小鼠深Ⅱ度烧伤的疗效可能与TNF-α、ICAM-1有关。
Objective To observe the curative effect and the effects of Shanlaizi oil dressing on tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule-1 (ICAM-1) in deep second degree burn mice. Methods Thirty mice were randomly divided into normal group, model group and shadaizi oil cataplasm group, 6 in normal group and 12 in the other two groups. After 4 h, intervention was given to observe the general indicators of burn. The mice were sacrificed in two batches. At 4 days, 6 mice were randomly sacrificed in the model group and the shanzuzi oil cataplasm group. After 12 days, the remaining mice were sacrificed and blood was collected from the eye to separate the burn tissue. The pathological changes of the skin tissue were observed by HE staining , Elisa and IHC were used to detect the expression of TNF-α and ICAM-1 in burn tissues. Results The wound healing area of Shadaizi Ointment on 4 d and 12 d was significantly smaller than that of the corresponding model group (P <0.05, P <0.01). Inflammatory cell infiltration decreased and all wounds of 12 mice healed on the 12th day. At 4 d and 12 d, the expression of ICAM-1 in shadzuzi ointment group was lower than that in model group (P <0.05). At 4 d, the expression of TNF-α in shadzuzi ointment group was lower than that in model group (P <0.05) On the 12th day, the shadzuzi oil dressing group was significantly higher than the model group (P <0.05). Conclusions Shanyukowu ointment has the functions of anti-inflammation, reducing scar and shortening the healing time. TNF-α can be reduced in acute phase, and its relative amount can be maintained in repair phase. Acute and reparative phase all reduce ICAM-1 expression. The efficacy of shadzuzi oil dressing on deep second degree burn in mice may be related to TNF-α and ICAM-1.
