本文用玻璃微电极细胞内记录方法观察了Co照射、平阳霉素和普鲁卡因对人低分化鼻咽癌细胞系单克隆株(B7)细胞膜电位的影响q结果显示:Co照射后4h膜电位负值减少,8~24h膜电位回升,48h时膜电位负值再次显著减少。不同剂量组48h膜电位差异非常显著(P<0.01),膜电位变化与照射剂量成正变关系;膜电位负值随平阳霉素作用时间延长而逐渐减小;平阳霉素合并Co照射使膜电位负值下降率大于二者分别作用之和(P<0.01);普鲁卡因作用后膜电位较早出现明显、不可逆转的下降。结果提示,细胞膜离子转运蛋白质可能参与平阳霉素、普鲁卡因抗肿瘤及Co照射杀伤肿瘤细胞的作用。
In this study, the effects of Co irradiation, pingyangmycin and procaine on the membrane potential of human low differentiated nasopharyngeal carcinoma cell line monoclonal strain (B7) were observed by glass microelectrode intracellular recording method. The results showed that after 4h Co irradiation Negative potential decreases, 8 ~ 24h membrane potential rise, 48h membrane potential negative again significantly reduced. The difference of membrane potential at 48h was significant (P <0.01), and the change of membrane potential was positively correlated with the irradiation dose. The negative value of membrane potential decreased with the prolonging of pingyangmycin time. Bleomycin combined with Co irradiation The decreasing rate of negative value of membrane potential was greater than the sum of the two effects (P <0.01). The membrane potential of procaine showed a significant and irreversible decline earlier. The results suggest that cell membrane ion transport proteins may be involved in pingyangmycin, procaine anti-tumor and Co irradiation kill tumor cells.
