目的研究芦荟大黄素对白细胞介素-2(IL-2)引起的T淋巴细胞增殖和细胞内Ca2+浓度([Ca2+]i)变化的影响,探讨芦荟大黄素对淋巴细胞增殖的作用和机制。方法采用MTT法检测细胞增殖,用单细胞钙成像系统记录细胞胞浆[Ca2+]i。结果芦荟大黄素对IL-2引起的T细胞增殖有显著抑制作用,且抑制效果与剂量有关;IL-2引起[Ca2+]i升高,并持续维持高[Ca2+]i水平,引起[Ca2+]i升高的机制包括胞内Ca2+释放和胞外Ca2+内流;芦荟大黄素显著抑制IL-2引起的[Ca2+]i升高,作用途径包括抑制胞内Ca2+释放和胞外Ca2+内流。结论芦荟大黄素对IL-2诱发的T细胞增殖有显著的抑制作用,其作用机制可能与抑制细胞内[Ca2+]i升高相关。
Objective To investigate the effect of aloe-emodin on the proliferation of T lymphocytes and the change of intracellular Ca2 + concentration induced by interleukin-2 (IL-2), and to explore the effect and mechanism of aloe-emodin on lymphocyte proliferation. Methods MTT assay was used to detect the proliferation of cells. Single cell calcium imaging system was used to record the cytoplasmic [Ca2 +] i. Results Aloe-emodin significantly inhibited the proliferation of T cells induced by IL-2, and the inhibitory effect was dose-dependent. IL-2 caused a rise of [Ca2 +] i and sustained high [Ca2 +] i levels, i increased the mechanism of intracellular Ca2 + release and extracellular Ca2 + influx; aloe-emodin significantly inhibited IL-2-induced [Ca2 +] i increased by the role, including inhibition of intracellular Ca2 release and extracellular Ca2 influx. Conclusions Aloe-emodin can significantly inhibit the proliferation of T cells induced by IL-2, and its mechanism may be related to the inhibition of intracellular [Ca2 +] i.
