目的:观察缺氧大鼠右心室心肌胶原网架改建的特点,并探讨血浆Ⅲ型前胶原( P CⅢ)含量的变化是否能反映心肌胶原含量和浓度的变化。方法:40 只 Wistar大鼠分为两组:①常氧对照组;②慢性缺氧组:缺氧大鼠暴露于摸拟5 000 m 高原30 d 和60 d,8 h/d。测定其 Pa O2,左、右心室收缩压,观察心肌胶原含量及形态学变化。结果:慢性缺氧可以引起 Pa O2 下降、右心室收缩压及右心室重量指数增加。缺氧大鼠右心室心肌胶原浓度和含量明显增加,而左心室则无明显变化。血浆 P CⅢ含量与心肌胶原浓度和含量呈正相关。病理切片检查显示缺氧大鼠右心室出现心肌内血管周围纤维化和局灶性纤维化。结论:慢性缺氧可引起右心室心肌胶原网架改建,血浆 P CⅢ含量的变化可反映右心室心肌胶原含量和浓度的变化。
OBJECTIVE: To observe the characteristics of remodeling of collagen network in right ventricular myocardium of hypoxia rats and to investigate whether changes of plasma procollagen Ⅲ (P Ⅲ) can reflect changes of myocardial collagen content and concentration. Methods: Forty Wistar rats were divided into two groups: ① normoxia control group; ② chronic hypoxia group: hypoxic rats were exposed to simulated 5000-m plateau for 30 days and 60 days for 8 hours. Pa Pa, left and right ventricular systolic pressure were measured to observe the myocardial collagen content and morphological changes. Results: Chronic hypoxia caused Pa O2 to decrease, right ventricular systolic pressure and right ventricular mass index to increase. Right ventricular myocardial collagen concentration and content increased significantly in hypoxic rats, while no significant changes in left ventricle. Plasma P C Ⅲ content and myocardial collagen concentration and content was positively correlated. Histopathological examination revealed perivascular fibrosis and focal fibrosis in the right ventricle of hypoxic rats. CONCLUSION: Chronic hypoxia can induce remodeling of collagen membrane in right ventricular myocardium. The change of plasma P CⅢ level can reflect the content and concentration of collagen in right ventricular myocardium.
