目的 研究IFN_α、TNF_α和IL_2联合激活正常骨髓细胞对白血病细胞的体外净化作用。方法 采用细胞集落形成法和逆转录多聚酶链反应 ,检测激活的正常骨髓对K562 细胞的净化作用。并观察其对正常造血及造血微环境的影响。结果 3种细胞因子均能激活骨髓细胞 ,以3种同时应用作用最强,且bcr/abl融合基因检测阴性 (Rt_PCR法 ) ;IL_2+IFN_α作用次之 ,bcr/abl融合基因阳性率41% ;其后是IL_2 +TNF_α和单独使用IL_2 ;最后是单用TNF -α ,bcr/abl融合基因均阳性。3种细胞因子对CFU_GM的抑制很小 ,对CFU_F和CFU_Blast有促进作用。结论 IL_2、IFN_α和TNF_α联合应用有助于净化白血病骨髓。
Objective To study the in vitro purification of leukemic cells by IFN-α, TNF_α and IL_2 combined with activation of normal bone marrow cells. METHODS: Colony formation and reverse transcription polymerase chain reaction were used to detect the purifying effect of activated normal bone marrow on K562 cells. And observe its effect on normal hematopoietic and hematopoietic microenvironment. Results All three cytokines could activate myeloid cells, and the three kinds of cytokines had the strongest effect at the same time. The detection of bcr/abl fusion gene was negative (Rt_PCR method). The effect of IL_2+IFN_α was the second, and the positive rate of bcr/abl fusion gene was 41%. Followed by IL2 + TNF_α and IL2 alone; Finally, with TNF-α alone, the bcr/abl fusion gene was positive. The inhibition of CFU_GM by the three cytokines was small, and it promoted CFU_F and CFU_Blast. Conclusion The combined use of IL-2, IFN-α and TNF_α can help to purify leukemia bone marrow.
