赤芍二基源药材的HPLC指纹图谱鉴别及质量评价

目的:通过HPLC指纹图谱研究,快速有效鉴别北赤芍与川赤芍,并评价其质量。方法:采用高效液相色谱法,分别以北赤芍与川赤芍对照药材为参照图谱,对不同产地、不同采收期的12批北赤芍与29批川赤芍进行指纹图谱研究,同时对其进行相似度评价,并采用判别分析,建立北赤芍与川赤芍的Fisher线性判别函数。结果:北赤芍对照药材与川赤芍对照药材的相似度约为0.7,<0.9,12批北赤芍与29批川赤芍与其相应对照药材的相似度都在0.9以上;北赤芍与川赤芍的Fisher线性判别函数为:Y北赤芍=139.797X1+21.961X2+30.927X3-14.796,Y川赤芍=45.688X1-10.867X2+69.898X3-13.440(自变量X1,X2,X3分别表示以芍药苷为参照峰的儿茶素、芍药内酯苷及苯甲酸的相对峰面积)。结论:采用HPLC指纹图谱,结合相似度评价或判别分析,可快速、有效鉴别北赤芍与川赤芍。川赤芍与北赤芍的化学成分既有共性,又存在差异,且川赤芍更复杂。物种来源不同是导致二者化学成分存在差异的主要因素,而产地及采收期对其影响不大,故应当根据上述研究并结合相关实验,制定规范的赤芍二基原药材质量标准,指导临床安全有效用药。 OBJECTIVE: To identify the quality of Petri red peony root and Radix Paeoniae Rubra fast and effectively by HPLC fingerprinting. Methods: The fingerprint of Radix Paeoniae Alba and Radix Paeoniae Alba was studied by HPLC, respectively. The fingerprints of 12 batches of Radix Paeoniae Rubra and 29 batches of Radix Paeoniae Alba were studied in different origins and recoveries. Simultaneously, The similarity was evaluated and discriminant analysis was used to establish the Fisher linear discriminant function of Radix Paeoniae and Radix Paeoniae Rubra. Results: The similarity between Radix Paeoniae Alba and Radix Paeoniae Alba is about 0.7, <0.9. The similarity of 12 batches of Radix Paeoniae Alba and Radix Paeoniae Alba is 29% Fisher’s linear discriminant function of Radix Paeoniae Alba is as follows: Y northern red peony root = 139.797X1 + 21.961X2 + 30.927X3-14.796, Y Chuan red peony root = 45.688X1-10.867X2 + 69.898X3-13.440 (independent variables X1, X2, X3 Indicates the relative peak area of ​​catechin, paeoniflorin and benzoic acid with paeoniflorin as the reference peak). Conclusion: HPLC fingerprinting, combined with similarity or discriminant analysis, can quickly and effectively distinguish between Radix Paeoniae and Radix Paeoniae Rubra. Chuan red peony and red peony root of the chemical composition of both common, but also differences, and Chuan red peony more complicated. Therefore, different sources of species are the main factors leading to differences in the chemical composition of the two raw materials, while the origin and harvest time have little effect on them. Therefore, according to the above research and related experiments, we should formulate normative quality standards of Radix Paecilomy Radix raw materials and guide Clinical safety and effective medication.
