目的采用分光光度法测定王不留行中的总皂苷。方法采用薄层色谱法分离总皂苷,对分光光度法的显色条件进行研究,并分析该方法的可行性。结果薄层色谱的展开剂为正丁醇-乙酸乙酯-水(4∶1∶5),显色反应的适宜条件为:5%香草醛-冰乙酸0.2 mL,高氯酸1.0 mL,70℃反应25 min;方法的线性范围为0.2~0.8 mg(r=0.9988);王不留行中总皂苷的含量为59.28%,加样回收率为101.27%,RSD=1.72%。结论所用方法稳定、可靠,可用于王不留行中总皂苷的含量测定和质量控制。
Objective To determine the total saponin in Wang Buyu line by spectrophotometry. Methods The total saponins were separated by TLC, the conditions of color development were studied by spectrophotometry, and the feasibility of the method was analyzed. Results The developing solvent of thin layer chromatography was n-butanol-ethyl acetate-water (4:1:5). The suitable conditions for the chromogenic reaction were 0.2% vanillin-glacial acetic acid, 1.0 mL perchloric acid, The linear range of the method was 0.2-0.8 mg (r = 0.9988). The content of total saponin in Wangbu stay was 59.28%. The recoveries were 101.27% and the RSD was 1.72%. Conclusion The method is stable and reliable, which can be used for the determination and quality control of total saponins in Wangbu stay line.
