通过比较了单层蔗糖分步离心和不连续蔗糖梯度离心制备的大鼠脑突触体对5-羟色胺和去甲肾上腺素的再摄取能力。结果表明,采用分步离心法可减少制备突触体的时间,降低操作难度,所得脑突触体具有有效蛋白含量较高、稳定性较好的优势。该法所制备的突触体对相应递质的特异性摄取能力有所提高。
The reuptake of serotonin and norepinephrine by rat brain synaptosomes prepared by comparing single-layered sucrose step-centrifugation and discontinuous sucrose gradient centrifugation was compared. The results showed that the step-by-step centrifugation can reduce the time for preparation of synaptosomes and reduce the difficulty of operation. The obtained synaptosomes have the advantages of higher effective protein content and better stability. The synaptosomes prepared by this method have higher specific uptake capacity for the corresponding transmitter.
