利用体外白蛋白非酶糖化系统,观察到体外白蛋白非酶糖化程度与温度、葡萄糖浓度、接触时间有明确关系。发现37℃、葡萄糖200mmol/L、4周为一周期的体外非酶糖化系统可以造成白蛋白的明显糖化,此系统可以作为体外白蛋白非酶糖化实验和药物筛选的理想体外非酶糖化系统。同时对一些中药或其成分进行的体外实验研究还表明:水飞蓟宾、槲皮素对体外白蛋白的非酶糖化有一定的抑制作用,IC_(50)值:对棕色反应产物分别为4.27×10~(-6)、2.36×10~(-6);对非酶糖化终产物(AGE)荧光各为7.30×10~(-6)、5.44×10~(-5)mol/L。
Using in vitro albumin non-enzymatic saccharification system, it was observed that the non-enzymatic saccharification of albumin in vitro has a clear relationship with temperature, glucose concentration, and contact time. It was found that in vitro non-enzymatic saccharification system at 37°C, glucose 200 mmol/L, and 4 weeks as a cycle can cause significant saccharification of albumin. This system can be used as an ideal non-enzymatic saccharification system for non-enzymatic saccharification experiments and drug screening in vitro. At the same time, some in vitro experiments conducted on some traditional Chinese medicines or their components also showed that silybin and quercetin have certain inhibitory effects on the non-enzymatic saccharification of albumin in vitro. The IC 50 value is 4 for brown reaction products, respectively. .27×10~(-6), 2.36×10~(-6); the fluorescence of non-enzymatic glycation end products (AGE) was 7.30×10~(-6), 5.44×10~ (-5) mol/L.
