目的研究组织蛋白酶D(Cath┐D)对乳腺癌侵袭、转移和预后的作用。方法采用免疫组织化学亲和素┐生物素┐过氧化物酶复合物(SABC)法,检测105例乳腺癌石蜡切片中肿瘤细胞(实质)和间质细胞组织蛋白酶D的表达,结合临床和病理资料分析其作用。结果组织蛋白酶D主要定位于细胞浆,部分增生的细胞可见定位于细胞核,间质细胞与实质细胞的Cath┐D的定位基本相似。间质细胞以肿瘤周围的间质细胞组织蛋白酶D的表达量为高。Cath┐D在乳腺癌实质和间质中总的表达率分别为49.5%(52/105),56.2%(59/105)。实质Cath┐D的表达与肿瘤细胞组织浸润、肿瘤大小、腋淋巴结转移、随访结果有关(P<0.05);与乳腺癌组织学分类、组织学分型、分级、年龄、ER、PR状态、局部复发、远处转移无关。间质Cath┐D的表达与乳腺癌肿瘤大小、腋淋巴结转移、局部复发、远处转移、年龄、ER、PR状态、局部复发无关。实质与间质Cath┐D的表达和乳腺癌患者的无瘤生存率有关(P<0.05);实质Cath┐D表达与总生存率无关(P>0.05),而间质的Cath┐D与总生存率有关(P<0.05)。结论实质和间质乳腺癌组织蛋白酶D的表达都和?
Objective To study the effect of cathepsin D (CathD) on invasion, metastasis and prognosis of breast cancer. Methods The immunohistochemical avidin-biotinylated peroxidase complex (SABC) method was used to detect the expression of cathepsin D in tumor cells (parenchyma) and mesenchymal cells in paraffin sections from 105 breast cancer patients. Data analysis of its role. Results Cathepsin D was mainly localized in the cytoplasm, and partially proliferated cells were localized in the nucleus. The localization of Cath D in the stromal cells and parenchymal cells was similar. In stromal cells, the expression of cathepsin D in the stromal cells around the tumor was high. The parenchyma was related to the expression of Cathepsin D and the tumor-free survival rate of breast cancer patients (P<0.05); the expression of Cathth D was not related to the overall survival (P>0.05), while the interstitial Cath ┐D was associated with overall survival (P<0.05). Conclusion The expression of cathepsin D in both parenchymal and interstitial breast cancers are both consistent.
