目的:探讨银杏叶提取物(GbE)对糖尿病大鼠肾脏保护作用及其机制。方法:将雄性SD大鼠建成链脲佐菌素诱导的糖尿病模型。设正常组(6只);糖尿病非治疗组(模型组,8只);糖尿病+银杏叶提取物治疗组(治疗组,8只)。6周后观察各组大鼠血糖、体质量、24h尿蛋白排泄、肾重等指标,并用免疫组织化学方法观察肾脏核因子-κB的表达及肾脏的病理形态学改变。结果:与模型组相比,治疗组肾脏肥大指数及24h尿白蛋白排泄量及肾组织血管紧张素Ⅱ含量明显降低(P<0.01,P<0.05),肾组织核因子-κB(NF-κB)及转化生长因子-β1(TGF-β1)的表达明显减少(P<0.05,P<0.01),同时糖尿病大鼠肾脏病理改变亦减轻。结论:银杏叶提取物通过下调核因子-κB及TGF-β1的表达发挥糖尿病肾病的保护作用。
Objective: To investigate the protective effects of Ginkgo biloba extract (GbE) on the kidney of diabetic rats and its mechanism. Methods: Male Sprague-Dawley rats were established as streptozotocin-induced diabetic model. The normal group (n = 6), diabetes non-treatment group (model group, n = 8) and diabetes plus ginkgo biloba extract treatment group (treatment group, n = 8). After 6 weeks, the indexes of blood glucose, body weight, excretion of urine protein and kidney weight of rats in each group were observed. The expression of nuclear factor-κB and pathological changes of kidney were observed by immunohistochemistry. Results: Compared with the model group, the renal hypertrophy index, 24h urinary albumin excretion and the content of angiotensin Ⅱ in renal tissue were significantly decreased (P <0.01, P <0.05), and the levels of nuclear factor-κB ) And TGF-β1 (P <0.05, P <0.01). At the same time, the renal pathological changes in diabetic rats were also alleviated. Conclusion: Ginkgo biloba extract exerts the protective effect of diabetic nephropathy by down-regulating the expression of nuclear factor-κB and TGF-β1.
