本文报告从北京郊区菜豆上分离到的一种小球状病毒。经鉴定是一种含单链DNA(ss-DNA)的病毒,称做菜豆畸矮病毒(Bean distortion dwarf virus,BDDV)。温室人工摩擦接种或白粉虱接种产生病株病状,表现为叶片畸形而变小,色深而质厚,有时带有轻微的花叶;严重时伴有褐色系统坏死,植株矮化,花果减少。寄主范围测定,在人工汁接的5科11种植物中只系统侵染菜豆和豇豆;由白粉虱接种的6科15种植物中除豆科外,还侵染曼陀罗、辣椒、烟草等茄科植物,但不发生局部侵染。提纯病毒颗粒的形态除部分为小球状呈散生外,多数呈晶格状聚集体,经Latex标准颗粒校正,每种颗粒直径均为9—11nm。从病叶组织超薄切片,观察到聚集在细胞质液泡附近呈晶格排列的病毒颗粒和在核膜周围、细胞质内散生的小球状病毒颗粒以及密集在叶绿体周围的成串病毒颗粒,大小为9—10nm。提纯的病毒核酸能被DNA酶I和S_1酶降解,而不被RNA酶A降解。提纯病毒回接在菜豆上具相当强的侵染性。
This article reports a small form of the virus isolated from beans in Beijing’s suburbs. It has been identified as a single-stranded DNA (ss-DNA) virus called Bean distortion dwarf virus (BDDV). Inoculation of artificial rubbing in greenhouse or inoculation of whitefly produced diseased plants, which manifested as leaf deformity becoming smaller, darker color and thicker, sometimes with slight mosaic; in severe cases brown system necrosis, dwarfing plants and decrease of flower and fruit . Host range determination, Artificial juice in 5 families and 11 kinds of plants in the systemic infection of beans and cowpea; by whitefly inoculation of 6 families of 15 kinds of plants in addition to leguminous crops, but also invasive Datura, pepper, tobacco, etc. Solanaceous plants, but no local infection. Purified virus particle morphology in addition to part of the pellet was scattered, most of the lattice-like aggregates, the Latex standard particles corrected, each particle diameter of 9-11nm. From the diseased leaves of the thin section, observed in the vicinity of the cytoplasmic vacuoles arranged in the lattice arrangement of virus particles and in the nuclear membrane around the cytoplasm of small spherical virus particles and dense clusters of viral particles around the chloroplasts, the size of 9-10 nm. The purified viral nucleic acid can be degraded by DNase I and S-1 enzymes without being degraded by RNase-A. Pure virus back in the relatively strong bean infectivity.
