目的 :研究经分离培养的兔骨髓基质干细胞 (MSC)接种在可吸收性纤维蛋白凝胶上向成骨细胞分化 ,表型维持的影响。方法 :取兔骨髓基质干细胞于含 10 0ml/L胎牛血清的DMEM培养液中培养 ,并向成骨细胞诱导。将细胞与纤维蛋白凝胶复合 ,通过扫描电镜观察细胞附着情况 ,共聚焦显微镜观察Ⅰ型胶原的分泌 ,组织化学染色检测碱性磷酸酶 (ALP)阳性细胞 ,ALP含量测定。结果 :骨髓基质干细胞在可吸收性纤维蛋白凝胶表面附着良好并继续增殖 ,可分泌Ⅰ型胶原 ,这些细胞的ALP染色呈强阳性 ,细胞碱性磷酸酶含量增高。结论 :具有良好的可塑性、低抗原性、好的降解吸收性的纤维蛋白凝胶具有促进骨髓基质细胞表达成骨细胞表型、合成细胞外基质的功能 ,是良好的细胞载体。
OBJECTIVE: To study the effect of isolated and cultured rabbit bone marrow stromal stem cells (MSC) on osteogenic differentiation and phenotype maintenance on absorbable fibrin gels. Methods: Rabbit bone marrow stromal cells were cultured in DMEM containing 100 ml / L fetal calf serum and induced to osteoblasts. The cells were mixed with fibrin gel, the cell adhesion was observed by scanning electron microscopy, the secretion of type I collagen was observed by confocal microscopy, and the alkaline phosphatase (ALP) positive cells and ALP were detected by histochemical staining. Results: BMSCs adhere well to the surface of absorbable fibrin gel and continue to proliferate, secreting type I collagen. The ALP staining of these cells was strongly positive and the content of alkaline phosphatase increased. CONCLUSION: Fibrin gel with good plasticity, low antigenicity and good biodegradability is a good cell carrier because it can promote bone marrow stromal cells to express osteoblast phenotype and synthesize extracellular matrix.
