以不同浓度的硫鱼精蛋白(PS)对Vero细胞制备的Ⅰ,Ⅱ,Ⅲ型脊髓灰质炎口服活疫苗(OPV)病毒悬液进行后处理去除细胞基质DNA研究.PS浓度大于1mg/mL,纯化后疫苗中DNA残余含量低于100pg/剂量.随着病毒液中PS浓度的增加,DNA残余含量减少,但病毒回收率亦降低.结果表明PS沉淀法,病毒回收率高,DNA残余含量能达到WHO规程DNA限量要求,对脊灰病毒rct/40特征及病毒壳蛋白等生物学性状无显著影响,是Vero细胞制备OPV简便的纯化方法
After treated with different concentration of protamine (PS), the virus suspension of oral live vaccine (OPV) of type I, II and III poliovirus prepared from Vero cells was treated to remove cell matrix DNA. PS concentration greater than 1mg / mL, purified DNA vaccine residual DNA content of less than 100pg / dose. As the concentration of PS in the virus increases, the residual DNA content decreases, but the virus recovery rate also decreases. The results showed that PS precipitation, high recovery rate of DNA, DNA residual content can meet the WHO regulatory DNA limit requirements, and have no significant effect on the rct / 40 characteristics of poliovirus and the biological properties such as the coat protein of poliovirus, which is a simple purification of OPV prepared by Vero cells method
