目的:观察灯盏花素对氯化钾诱导的培养的家兔平滑肌细胞内游离钙的影响。方法:用RF_5000荧光光度仪检测Fura-2AM负载的培养平滑肌细胞内游离钙的浓度。结果:在有细胞外钙存在时,静息条件下细胞内游离钙为330±100nmol·L-1,灯盏花素能轻度降低静息状态下细胞内游离钙,但无统计学意义。氯化钾50mmol·L-1将细胞内游离钙增加到420±104nmol·L-1,而灯盏花素10-6、10-5、3×10-5mol·L-1能进一步加强由氯化钾引起的细胞内游离钙的增加。然而,在无细胞外钙时,灯盏花素对细胞内游离钙却没有影响。结论:灯盏花素通过电压依赖性钙通道加强钙离子内流。
Objective: To observe the effect of erigeron breviscapus on the intracellular free calcium in cultured rabbit smooth muscle cells induced by potassium chloride. METHODS: The concentration of free calcium in cultured smooth muscle cells was measured by Fura-2AM using RF_5000 fluorescence spectrophotometer. RESULTS: In the presence of extracellular calcium, intracellular free calcium was 330±100 nmol·L-1 at rest, and breviscapin could slightly reduce intracellular free calcium in the resting state, but it was not statistically significant. Potassium chloride 50mmol·L-1 increased intracellular free calcium to 420±104nmol·L-1, while breviscapine 10-6, 10-5, 3×10-5mol·L-1 could be further enhanced by chlorination. Potassium-induced increase in intracellular free calcium. However, breviscapine had no effect on intracellular free calcium in the absence of extracellular calcium. Conclusion: Breviscapine enhances calcium influx through voltage-dependent calcium channels.
