目的:观察宫瘤消胶囊对子宫内膜异位症(内异症)模型大鼠异位内膜中凋亡基因Bcl-2、Bax蛋白的表达的影响,为该药治疗内异症提供理论依据。方法:于大鼠动情期行自体移植法建立子宫内膜异位症大鼠模型,造模成功后随机分为模型组、桂枝茯苓胶囊组及宫瘤消胶囊小、中、大剂量组,各组分别给予指定药物进行灌胃干预,另取一组正常大鼠为正常内膜组,用生理盐水灌胃。用免疫组织化学法检测各组大鼠异位内膜中凋亡基因Bcl-2和Bax的蛋白表达情况。结果:与正常内膜组比较,模型组Bax蛋白表达明显降低,Bcl-2蛋白的表达明显升高,Bax/Bcl-2比值降低(P<0.01);与模型组比较,宫瘤消胶囊各剂量组Bax蛋白表达明显升高,Bcl-2蛋白表达明显降低,Bax/Bcl-2比值增加(P<0.05,P<0.01),且作用随药物剂量增加而增加(P<0.05,P<0.01)。结论:宫瘤消胶囊可能通过调节凋亡基因Bcl-2和Bax的表达来实现对异位内膜的凋亡的控制。
Objective: To observe the effect of Gong-Yun-Xiao Capsule on the expression of Bcl-2 and Bax protein in the ectopic endometrium of endometriosis model rats and to provide theory for the treatment of endometriosis in accordance with. Methods: Rat model of endometriosis was established by autograft in estrus rats. After successful modeling, rats were randomly divided into model group, Guizhi Fuling capsule group and Gongyuxiao Capsule small, medium and large dose groups, Each group were assigned to the specified drug gavage intervention, the other group of normal rats as normal intima group, with saline gavage. Immunohistochemistry was used to detect the protein expression of apoptosis gene Bcl-2 and Bax in ectopic endometrium of each group. Results: Compared with the normal endometrium group, the expression of Bax protein and Bcl-2 protein in the model group were significantly lower than those in the normal endometrium group (P <0.01). Compared with the model group, (P <0.05, P <0.01), and the effect increased with the dose of Bax (P <0.05, P <0.01), and the Bcl-2 protein expression was significantly decreased ). Conclusion: Gongyuxiao Capsule may control the apoptosis of ectopic endometrium by regulating the expression of Bcl-2 and Bax.
