目的:研究荷S180瘤小鼠灌服中药复方抑瘤饮不同时间点肿瘤内血管生成的动态变化,揭示抑瘤饮体内抗瘤的作用机制。方法:56只BALB/c小鼠随机分为抑瘤饮组和对照组(n=28),每组分为4个亚组(每亚组7只)。右腋皮下接种S180肿瘤细胞24 h后,抑瘤饮组小鼠灌服抑瘤饮,对照组小鼠灌服等量凉开水,每日2次,每次0.5 mL;2组分别于灌服第10,20,30,40天点各处死一亚组小鼠。分离肿瘤制成石蜡包埋切片,免疫组化法检测肿瘤组织内血管生成(CD34染色)、血管内皮生长因子(VEGF)、血管内皮生长因子受体-2(VEGFR-2)和内皮抑素(ES)的动态变化,结果以阳性酶点(positive enzyme dot,PED)表示。结果:抑瘤饮能显著降低肿瘤组织内微血管生成:抑瘤饮组CD34在第10,20,30,40天点PED均低于对照组(分别P<0.05,P<0.01,P<0.01和P<0.01)。抑制VEGF和VEGFR-2的表达:抑瘤饮组VEGF在第10,20,30,40天点PED均低于对照组(分别P<0.05,P<0.01,P<0.01和P<0.01):抑瘤饮组VEGFR-2在相应时间点均低于对照组(分别P<0.05,P<0.01,P<0.01,P<0.01)。促进ES的表达:抑瘤饮组ES在相应时间点与对照组比第10天点PED无明显差异,在第20,30,40天点PED均高于对照组(分别P<0.05,P<0.05和P<0.01)。结论:抑瘤饮体内可通过下调VEGF和VEGFR-2表达,上调ES表达,抑制肿瘤组织内血管生成发挥抗瘤作用。
OBJECTIVE: To study the dynamic changes of tumor angiogenesis at different time points in the S180 tumor-bearing mice fed with traditional Chinese medicine SJY and to reveal the anti-tumor mechanism of SQYD in vivo. METHODS: Fifty-six BALB/c mice were randomly divided into two groups, the Yiliu Yin group and the control group (n=28). Each group contained 4 subgroups (7 rats in each subgroup). After S180 tumor cells were inoculated subcutaneously in the right eye for 24 h, mice in the Yiliuyin group were treated with Yiliuyin; mice in the control group were given the same amount of cold water twice a day, 0.5 mL each time; 2 groups were administered separately. On the 10th, 20th, 30th and 40th days, a subgroup of mice was sacrificed. The tumors were isolated and paraffin-embedded sections were made. The angiogenesis (CD34 staining), vascular endothelial growth factor (VEGF), vascular endothelial growth factor receptor-2 (VEGFR-2) and endostatin were detected by immunohistochemistry. Dynamic changes in ES) are indicated as positive enzyme dots (PED). RESULTS: SQYD could significantly reduce the microvessel production in tumor tissues: PED in the Yishu Yin group was lower than that in the control group at the 10th, 20th, 30th, and 40th days (P<0.05, P<0.01, P<0.01, respectively).
