为了建立磷酸己糖异构酶(PHI) 的酶偶联测定法, 并观察其临床应用价值, 根据PHI可催化6-磷酸果糖转化为6-磷酸葡萄糖(G6P), 在6-磷酸葡萄糖脱氢酶催化下, 6-磷酸葡萄糖转化为6-磷酸葡萄糖酸, 同时使NADP还原为NADPH, 在340 nm 处吸光度变化速率与PHI的活性成正比。方法的批内CV< 1.2% , 批间CV< 1.5% , 回收率98% ~102% , 线性达1000 U/L。健康对照组PHI95% 可信限为20~89 U/L。取PHI150 U/L作为肺癌与其他肺部疾病鉴别诊断指标, 鉴别诊断效率达0.805。
In order to establish an enzyme conjugate assay for phosphohexose isomerase (PHI) and observe its clinical application value, PHI can catalyze the conversion of 6-phosphorylated fructose to glucose-6-phosphate (G6P), which dehydrogenates glucose at 6-phosphate. Enzymatically, glucose 6-phosphate is converted to 6-phosphogluconate and NADP is reduced to NADPH. The rate of change in absorbance at 340 nm is proportional to the activity of PHI. The method has a batch CV <1.2%, an inter-assay CV < 1.5%, a recovery of 98% ~ 102%, and a linearity of 1000 U/L. The PHI 95% confidence limit of the healthy control group was 20-89 U/L. PHI150 U/L was used as a differential diagnosis index for lung cancer and other lung diseases. The differential diagnosis efficiency was 0.805.
