目的:研究天山雪莲主要活性成分绿原酸和芦丁的大鼠在体肠吸收动力学。方法:采用单向灌流实验技术,利用HPLC法测定绿原酸和芦丁的量,分别研究药物质量浓度及吸收部位对绿原酸和芦丁吸收的影响。结果:药物质量浓度对绿原酸和芦丁吸收速率常数(Ka)和表观吸收系数(Papp)无显著性影响;绿原酸和芦丁在小肠各段间吸收的Ka和Papp无显著性差异,但与结肠相比吸收明显增大。结论:一定范围的药物浓度对绿原酸和芦丁的Ka和Papp无显著性影响,其吸收机制为被动扩散。药物在全肠道吸收较好,吸收窗主要在小肠,且小肠内无明显的特定吸收部位。
OBJECTIVE: To study the in vivo intestinal absorption kinetics of chlorogenic acid and rutin, the main active components of snowball in Tianshan Mountains. Methods: Uni-directional perfusion technique was used to determine the amount of chlorogenic acid and rutin by HPLC. The effects of drug concentration and absorbed site on the absorption of chlorogenic acid and rutin were studied. Results: There was no significant difference in Ka (superscript +) and apparent absorption coefficient (Papp) between chlorogenic acid and rutin, and Ka and Papp between chlorogenic acid and rutin Difference, but significantly increased compared with the colon absorption. Conclusion: A certain range of drug concentration has no significant effect on Ka and Papp of chlorogenic acid and rutin, and its absorption mechanism is passive diffusion. Drug absorption in the whole intestine is good, the absorption window is mainly in the small intestine, and there is no obvious specific absorption site in the small intestine.
