目的探讨胚胎绒毛组织中胰岛素样生长因子-2(IGF-Ⅱ)和H19印记基因表达与过期流产的关系。方法应用逆转录多聚酶链反应(RT-PCR)技术测定正常早孕女性38例(对照组)和41例过期流产患者(实验组)胚胎绒毛组织中IGF-Ⅱ和H19印记基因的表达水平,并进行对比分析。结果实验组胚胎绒毛组织中IGF-ⅡmRNA的表达显著低于对照组(t=8.662,P<0.05);H19 mRNA的表达显著高于对照组(t=12.706,P<0.05)。结论胚胎绒毛组织中IGF-Ⅱ、H19印记基因的异常表达可影响胚胎的正常发育,可能是导致过期流产发生的原因之一,可为过期流产的基因诊断提供新的切入点。
Objective To investigate the relationship between the expression of insulin-like growth factor-2 (IGF-Ⅱ) and H19 imprinting genes in embryonic villi and overdue abortion. Methods RT-PCR was used to detect the expression of IGF-Ⅱ and H19 in the chorionic tissue of 38 normal pregnant women (control group) and 41 abortion patients (experimental group) Comparative analysis. Results The expression of IGF-ⅡmRNA in the experimental group was significantly lower than that in the control group (t = 8.662, P <0.05). The expression of H19 mRNA in the experimental group was significantly higher than that in the control group (t = 12.706, P <0.05). Conclusion Abnormal expression of IGF-Ⅱ and H19 imprinted genes in embryonic villi may affect the normal development of embryos and may be one of the causes of over-abortion. It may provide a new entry point for genetic diagnosis of abortion.
