目的:观察强筋健骨胶囊对佐剂关节炎(AA)大鼠血清IL-6、CRP、RA33、IgM-RF、IgA、IgG水平的影响并探讨其治疗类风湿性关节炎的作用机理。方法:将120只雄性SD大鼠随机分为正常对照组、模型对照组、夏天无胶囊组、白芍总苷(TGP)胶囊组、甲氨蝶呤(MTX)组、强筋健骨胶囊高、中、低剂量组,每组15只。除正常对照组外其余各组大鼠右后足趾皮内注射0.1mL FCA致炎并诱发大鼠佐剂关节炎,对照组大鼠注射同等剂量的生理盐水。按给药方案各干预组造模前预防性给药4d,造模后给药治疗21d,实验过程中按方案要求测量各组大鼠体质量、足趾周长、关节炎指数(AI);治疗结束后颈总动脉取血,采用酶联免疫吸附法(ELISA)检测大鼠血清IL-6、CRP、RA33、IgM-RF、IgA、IgG的含量。结果:与正常对照组比:模型对照组大鼠血清IL-6、CRP、IgG、IgA,足趾肿胀度、AI均显著升高(P<0.05,P<0.01);IgM-RF、RA33呈升高趋势(P>0.05)。与模型对照组相比:强筋健骨胶囊各剂量组足趾肿胀度、AI、IL-6、CRP、IgG、IgA均显著降低(P<0.05,P<0.01)。与其它各治疗组相比:强筋健骨胶囊小剂量组足趾肿胀度显著降低(P<0.01),大剂量组AI极显著降低(P<0.01);IL-6、CRP、IgG、IgA含量显著降低(P<0.05,P<0.01);IgM-RF、RA33水平呈下降趋势(P>0.05);MTX组体质量明显降低(P<0.01)。结论:强筋健骨胶囊显著降低AA模型大鼠足趾肿胀度和关节炎指数,下调血清IL-6、CRP、IgG、IgA水平,效应强度优于夏天无胶囊、白芍总苷和甲氨蝶呤,效应机理系通过下调前致炎因子IL-6所诱导的细胞、体液超敏反应。
OBJECTIVE: To observe the effect of QJG on IL-6, CRP, RA33, IgM-RF, IgA and IgG in serum of adjuvant arthritis (AA) rats and to explore its mechanism of action in treating rheumatoid arthritis. Methods: 120 male Sprague-Dawley rats were randomly divided into normal control group, model control group, summer no capsule group, TGP capsule group, methotrexate (MTX) group and Jiangqijiang capsule , Medium and low dose group, each group 15. In addition to the normal control group, rats in other groups were intradermally injected with 0.1 mL FCA into the right hind foot to induce arthritis in rats, and rats in the control group were injected with the same dose of saline. According to the protocol of prescription, the body weight, toe circumference and arthritis index (AI) of rats in each intervention group were measured prophylactically for 4 days and 21 days after modeling. After the treatment, the common carotid arteries were taken for blood. The contents of IL-6, CRP, RA33, IgM-RF, IgA and IgG in the serum were detected by enzyme-linked immunosorbent assay (ELISA) Results: The levels of IL-6, CRP, IgG, IgA, toe swelling and AI in model control group were significantly higher than those in normal control group (P <0.05, P <0.01) Increasing trend (P> 0.05). Compared with the model control group, the Qiangjianjiangyu capsule significantly reduced the degree of toe swelling, AI, IL-6, CRP, IgG and IgA in all dose groups (P <0.05, P <0.01). Compared with other treatment groups, the Qiangqijianggu capsule low dose group significantly reduced the degree of toe swelling (P <0.01), high dose group AI was significantly lower (P <0.01), IL-6, CRP, IgG, IgA (P <0.05, P <0.01). The levels of IgM-RF and RA33 showed a decreasing trend (P> 0.05). The body weight of MTX group was significantly lower (P <0.01). CONCLUSION: Qiangqijianggu Capsule can significantly reduce the degree of toe swelling and arthritis index, decrease the levels of IL-6, CRP, IgG and IgA in AA model rats. The effect intensity is better than that of summer without capsule, Pterin, the mechanism of action through the down-regulation of inflammatory cytokines IL-6-induced cell, humoral hypersensitivity.
