目的 应用原子力显微镜 (AFM)实时观察体外培养的内皮细胞膜表面超微结构 ,并观察氧化低密度脂蛋白 (oxLDL)的影响。方法 以体外培养的大鼠脑微血管内皮细胞为材料 ,用AFM观察 80 μg/mloxLDL对细胞膜的影响。 结果 AFM可以观察内皮细胞表面纳米超微结构 ,oxLDL可引起超微结构改变。结论 首次用AFM观察oxLDL对微血管内皮膜的影响 ,AFM在高脂血症实验研究中有应用前景。
Objective To observe the ultrastructure of endothelial cell membrane in vitro by atomic force microscope (AFM) and observe the effect of oxLDL. Methods The rat cerebral microvascular endothelial cells cultured in vitro were used as materials to observe the effect of 80 μg / ml xLDL on the cell membrane by AFM. Results AFM could observe the ultrastructure of endothelial cells and oxLDL caused ultrastructural changes. Conclusions For the first time, the effect of oxLDL on the microvessel endothelium was observed by AFM. AFM has the potential application in the experimental study of hyperlipidemia.
