猪圆环病毒2型(PCV-2)严重制约着养猪业的发展,目前尚无针对该病的有效疫苗和药物。为了加强PCV-2的监控及预防,根据Gen Bank中已发布的PCV-2的序列设计了一对特异性引物,对PCR的反应条件进行优化,构建了PCV-2的PCR检测方法,并进行了特异性检测和敏感性试验,并用该方法对泰州地区67份疑似病料进行了检测。结果表明:建立的PCR方法操作简单,技术要求较低,便于临床检测或流行病学调查,所检测的泰州地区67份疑似断奶仔猪多系统衰竭综合征(PMWS)的病料(淋巴结),其中42份表现为阳性,阳性率可达62.9%。说明该检测方法的建立有利于规模化猪场对PCV-2的检测及监控,可为该病的早期诊断提供技术平台。
Porcine circovirus type 2 (PCV-2) severely restricts the development of swine industry, there is no effective vaccine against the disease and drugs. In order to strengthen the monitoring and prevention of PCV-2, a pair of specific primers was designed according to the published sequence of PCV-2 in Gen Bank to optimize the reaction conditions of PCR, and the PCR detection method of PCV-2 was constructed. The specificity test and the sensitivity test were carried out, and 67 suspected cases in Taizhou area were tested by this method. The results showed that the established PCR method was simple, less technically demanding and convenient for clinical or epidemiological investigation. The data of 67 cases of suspected weaned piglets with multiple system failure syndrome (PMWS) in Taizhou area were detected, including 42 were positive, the positive rate of up to 62.9%. It is indicated that the establishment of this detection method is beneficial to the detection and monitoring of PCV-2 in large-scale pig farms and provides a technological platform for the early diagnosis of this disease.
