目的探讨溶质相关载体26A3(SLC26A3)在大鼠动情周期子宫及卵巢内的表达。方法根据阴道涂片检测结果,将12只雌性Wister大鼠分别在动情前期(P)、动情期(E)、动情后期(M)和动情间期(D)断头处死,取出卵巢及子宫,提取总RNA及总蛋白,用RT-PCR及Western blot方法检测卵巢及子宫内SLC26A3在动情周期中各时期的表达。结果子宫内SLC26A3在动情期mRNA的转录水平和蛋白的表达水平相对较高,卵巢内SLC26A3在动情前期和动情期mRNA的转录水平和蛋白的表达水平相对较高。结论动情周期中,SLC26A3在大鼠卵巢及子宫内呈规律性表达,其变化为受孕做好了充分准备。
Objective To investigate the expression of solute-associated carrier 26A3 (SLC26A3) in uterus and ovary during estrous cycle in rats. Methods According to the result of vaginal smear test, 12 female Wister rats were sacrificed at the early estrus (P), estrus (E), late estrus (M) and estrus (D) Total RNA and total protein were extracted. The expression of SLC26A3 in ovary and uterus during estrous cycle was detected by RT-PCR and Western blot. Results Intrauterine mRNA and protein expression of SLC26A3 in estrous phase were relatively high. The mRNA and protein expression of SLC26A3 in ovary were relatively higher in pre-estrus and estrus phases. Conclusion During the estrous cycle, SLC26A3 is regularly expressed in the ovary and uterus of the rats, and the changes are well prepared for pregnancy.
