目的:研究基质金属蛋白酶(matrix metalloproteinases,MMP)MMP2、MMP7在甲状腺滤泡上皮性肿瘤中表达及其与病变的关系。方法:应用免疫组化及原位杂交方法检测78例甲状腺滤泡上皮性肿瘤(腺癌38例,腺瘤40例)标本中MMP2、MMP7表达。结果:MMP2及MMP7蛋白在各甲状腺肿瘤组织表达中,腺癌表达显著高于腺瘤(P<0.01);MMP2及MMP7蛋白表达与腺癌转移有相关性(P<0.05),肿瘤间MMP2及MMP7的 mRNA与蛋白表达呈正相关性(P<0.01)。结论:MMP2、MMP7的表达与甲状腺肿瘤显著相关,它们在甲状腺癌病变中高度表达,检测MMP2及MMP7蛋白或 mRNA水平对于鉴别甲状腺滤泡上皮性肿瘤良恶性具有重要参考意义。
Objective: To investigate the expression of matrix metalloproteinases (MMPs) MMP2 and MMP7 in thyroid follicular epithelial tumors and its relationship with lesions. Methods: Immunohistochemistry and in situ hybridization were used to detect the expression of MMP2 and MMP7 in 78 cases of thyroid follicular epithelial tumor (38 cases of adenocarcinoma and 40 cases of adenoma). Results: The expressions of MMP2 and MMP7 in adenocarcinoma were significantly higher than those in adenoma (P <0.01). The expressions of MMP2 and MMP7 were correlated with the metastasis of adenocarcinoma (P <0.05) MMP7 mRNA and protein expression was positively correlated (P <0.01). Conclusion: The expressions of MMP2 and MMP7 are significantly correlated with thyroid tumors. They are highly expressed in thyroid carcinomas. Detecting the levels of MMP2 and MMP7 protein or mRNA may be useful for differentiating thyroid follicular epithelial tumors from malignant ones.
