目的 :研究神经细胞经缺氧 /缺糖培养诱导的凋亡发生率、胞内游离钙离子变化及参麦注射液的治疗作用。方法 :碘化丙啶染色 ,流式细胞仪定量分析细胞凋亡百分率 ;Fluo 3负载 ,流式细胞仪观察胞内钙平均荧光值变化。结果 :缺氧 /缺糖培养可诱导神经细胞凋亡和引起细胞内游离钙离子增高 ,参麦注射液能抑制细胞凋亡、降低细胞内游离钙离子浓度。结论 :凋亡是神经细胞死亡方式之一 ,参麦注射液能抑制缺氧 /缺糖诱导的神经细胞凋亡及钙超载 ,有神经细胞保护作用。
Objective : To study the rate of apoptosis induced by hypoxia/glucose deprivation in cultured neurons, intracellular free calcium ion, and the therapeutic effect of Shenmai injection. Methods: propidium iodide staining, flow cytometry quantitative analysis of apoptosis percentage; Fluo 3 load, flow cytometry observed intracellular calcium fluorescence changes. RESULTS: Hypoxia/glucose deprivation could induce neuronal apoptosis and increase intracellular free calcium ion. Shenmai injection could inhibit apoptosis and reduce intracellular free calcium concentration. Conclusion: Apoptosis is one of the ways of neuronal cell death. Shenmai injection can inhibit neuronal apoptosis and calcium overload induced by hypoxia/glucose deprivation and has neuroprotective effect.
